hardcore mode involves harder style rankings, tweaked combos, and modified devil trigger, so while you might want to do that on a separate run, you can run it on your first playthrough if you really want to speedily complete the game.
If you are good at DMC or the hack and slash genre it is recommended to start on Nephilim, since you will need to do a playthrough on hardcore mode on Nephilim or above for both campaigns. Human is easy, Devil hunter is normal, and Nephilim is hard. There are three to start with: human, devil hunter, and Nephilim. If you are a DMC veteran or have played the original version, you will know where you want to start, but if you are just trying it out, you will want to watch which difficulty you choose. Where you start in DMC all depends on your prior experience. However, in order to get all of his upgrades and collectibles you will also need quite a few runs. Not only will you have to do a couple of playthroughs for Dante, but Vergil also gets his turn in the spotlight with the included Vergil's Downfall DLC – and luckily the requirements are much easier. That being said, multiple playthroughs will be more than welcomed as you will need to do at least one to gain access to all of the abilities and weapons, which will in turn give you access to all of the collectibles and secret missions. Devil May Cry is all about those playthroughs – you can't hope to get more than half of the gamerscore (or even half of the upgrades) on a single playthrough, even on Nephilim. Luckily only hardcore has achievements tied to it.
